can u tell me what is the interaction between the sheep on the 2 river's tiles with the game?

The shepherd scores the sheep tokens plus the sheep on the river tile.

Yes, this is permanent sheep.

Can I score just the 2 sheep on those tiles with my sheperd, or do I need to add more ?

As soon as you pull the shepherd you need to draw a sheep.. so.. you already have 1 more. and the next time you add field you can pull it back

Rules for the shepherd assume to place a shepherd, draw a token and only after the next move you can score the flock (if you expand the shepherd field, of course). If you draw a wolf right after placing the shepherd, you can't score anything, even those two "neutral" sheep.
